The CD45RA, also called Leukocyte Common Antigen, is a sialoglycoprotein membrane with three different exons that encode the peptide segments, which are designated as A, B, and C. The various splicing options of the exons will provide a variety of isoforms. It is considered a single-chain glycoprotein and is expressed in 40 to 50 percent of all peripheral CD4+ T-cells, 50 percent of all B-cells, Leukemic B-cell lines, and CD8+ T-cells. The T-cells for CD45RA and CD45RO can define complementary non-overlapping populations of the resting peripheral T-cells. It is considered a virgin or naïve T-cell.
More Information About This Antibody
This antibody is designed for research use and isn’t available for diagnostic procedures. It does have a clone called SPM504, and its immunogen is the stimulated human leucocytes. The epitope is not determined, but the IG isotype is IgG(2a). Its molecular weight is between 205 and 220 kDa.
CD45RA has been tested on humans.
Applications And Procedure
It is designed for Immunohistochemistry (IHC) applications. The procedure for IHC is to prepare the specimen using Formalin-fixed tissues embedded in paraffin. Deparaffinized slides are required using graded alcohols or xylene. When using the concentrated format of the product, you will dilute this antibody in a 1:200 ratio. Dilutions are estimations. Your results may be different based on various protocols and methods. There is no antigen retrieval and incubation periods are for 30 minutes while sitting at room temperature.
You should rinse the slides between use with PBS/0.05 percent Tween. The positive control for CD45RA is the tonsil, and the cellular localization is the membrane.
This product should be stored between two and eight degrees Celsius.
